شناسایی مولکولی تنوع آللی در مکان‌های ژنی VRN-1 ،و ,VRN-B3 کنترل کننده نیاز بهاره‌سازی در ژنوتیپ‌های گندم بر مبنای چندشکلی موجود در پروموتر و نواحی اینترون

چکیده

گندم، یکی از گیاهان مهم زراعی ایران و جهان، از جنبه‌های گوناگون غذایی و اقتصادی حائز اهمیت می‌باشد. شناسائی تنوع آللی ژن‌های کنترل کننده نیاز بهاره‌سازی در ارقام و لاین‌های مختلف گندم، امکان تعیین تیپ رشد، پیش‌بینی سطح تحمل سرما و سازگاری ژنوتیپ‌های مختلف گندم را به اقلیم‌ها و مناطق هدف فراهم می‌سازد. در سطح مولکولی طول دوره ورنالیزاسیون در ارقام گندم به طور عمده به وسیله مکان‌های ژنی (VRN-1، VRN-2وVRN-3) تعیین می­شود. در گندم هگزاپلویید ژن‌های( Vrn) برای تیپ رشد بهاره در فرم غالب و برای تیپ زمستانه در فرم مغلوب همراه با اثرات اپی­استاتیک عمل می‌کنند. در این تحقیق تنوع آللی موجود در مکان‌های ژنی (VRN-1 و VRN-B3) در 50 ژنوتیپ گندم نان با استفاده از 9 جفت آغازگر اختصاصی تعیین شد. آلل‌های غالب  Vrn-B1,Vrn-D1)و Vrn-B3) به­ترتیب با فراوانی (٪58، ٪22 و ٪90( شناسایی شدند. همچنین آلل‌های مغلوب vrn-B1, vrn-D1vrn-B3)  به­ترتیب با فراوانی (٪42، ٪78 و ٪10) شناسایی شدند. در مکان ژنی (VRN-A1) آلل Vrn-A1c)) با فراوانی 58٪، آلل (Vrn-A1b) با فراوانی26٪، آلل (Vrn-A1a) با فراوانی14٪ و آلل (vrn-A1) با فراوانی 2٪ دارای کمترین فراوانی بود. این تحقیق تنوّع اللی قابل ملاحظه‌ای را در مکان‌های ژنی کنترل کننده ورنالیزاسیون و تحمل سرما در ژنوتیپ­های گندم دیم مورد بررسی نشان داد که اطلاعات حاصل در جهت تجمیع الل‌های مطلوب برای تولید لاین‌های سازگار به شرایط اقلیمی در مناطق هدف مورد بهره‌برداری قرار خواهد گرفت.

Wheat, as a major crop in Iran and in the world, is considered an important crop for human consumption. Identification of allelic variations at loci that control vernalization requirements and cold tolerance, provides valuable information for determining adaptability of wheat genotypes to target environments. At molecular level, vernalization in wheat (Triticum aestivum L.) is determined mainly by three VRN-1, VRN-2 and VRN-3 loci. In hexaploid wheat, dominant alleles Vrn-A1, Vrn-B1 and Vrn-D1 are responsible for spring growth habit and recessive allels with epistatic interactions determine the winter growth habit. In this study, allelic variations among 50 wheat genotypes were studied using 9 primer pairs specific to VRN-1 and VRN-B3 loci. Dominant alleles at Vrn-B1,Vrn-D1 and Vrn-B3 loci were detected with the frequency of %58, %24 and %90, respectively. Recessive alleles vrn-B1, vrn-D1 and vrn-B3 had a frequency of %42, %76 and %10, respectively. At Vrn-A1 locus, different alleles were detected with the frequency of Vrn-A1c (58%), Vrn-A1b (26%), Vrn-A1a (14%) and vrn-A1 (2%). Our finding revealed a considerable level of variations at gene loci controlling vernalization and cold tolerance in dryland wheat genotypes. Data provided here may be used for producing new wheat cultivars adapted to the target environments.
